No. As long as you don't remove the SPF 55 sunblock when applying your makeup, you will still have the benefit of the SPF 55 sunblock. It is important to remember, however, that you have to reapply sunscreen or you will not receive its full benefit.
Even products advertised as waterproof eventually wash off or sweat off. You must reapply the product to receive the SPF protection promised. The American Academy of Dermatology recommends applying sunscreens to dry skin 15-30 minutes before going outdoors and re-applying it at least every two hours or after swimming or sweating heavily.
Be sure to apply enough sunscreen to cover exposed areas. Remember to apply sunscreen even on cloudy days. And wear protective clothing if you are out in the sun for any length of time.
